The direct powder tablet pill pressing process has high requirements on the properties of raw materials and excipients, and the choice of excipients should usually be fully studied before product development. 

Common excipients for powder direct compression, filling excipients include various types of microcrystalline cellulose, lactose, calcium hydrogen phosphate dihydrate, compressible starch, micropowder silica gel, etc.;


The widely used disintegrants include sodium starch glycolate, low-substituted hydroxypropyl cellulose, etc., but polyvinylpyrrolidone, which is the three major components of disintegrants, is not suitable as a tablet disintegrant under direct powder compression technology.Because of its strong hygroscopicity, which will often cause cracking on the surface of the tablet during tablet storage.

In addition, in the process of development and production, the brittleness of some varieties would make it difficult to meet the production requirements. In order to reduce the brittleness, a small amount of adhesives will be added to the formulation. These adhesives should have weak hygroscopicity, good fluidity and plasticity. and other characteristics, including hydroxypropyl cellulose and povidone with high adhesive force and low solution viscosity. Filling excipients are the main excipients for tablets, with the largest amount and the largest proportion, which can most affect the fluidity and compressibility of excipients, and even affect drug dissolution.

Good fluidity is the prerequisite and basis for good mixing uniformity. Good compressibility can reduce tablet friability, difficulty in forming, and reduce the risk of mechanical properties to tablets.
